I think is hilarious that you have The Island on the list :D. It was laughably bad!

Close call btwn Wedding Crashers and 40yr Old Virgin. All I have to do is think about the scene at the dinner table with Vince Vaughn and it wins the vote!

It was a toss up between Kung Fu Hustle and 40 Year old Virgin. KFH was the eventual winner for me because it was so unexpected and fresh. Sheeeit I know some 40 plus year old virgins, both men and women and I find little funny about them, but the body waxing scene and the extended version was downright hilarious.

Waiting was pretty funny. Underrated movie. Even my mom thought it was funny.

I noticed that you put Me & You & Everyone We Know in the drama category, but I don't think it's much of a stretch to call it an "existensial comedy." It was definitely the movie that I laughed hardest at this year. "Back and forth. Forever." The Squid & the Whale also blurred the line between drama and comedy-- you know what, what great movie doesn't have moments of sublime humor, like life?-- Jeff Daniels' performance in particular was both hilarious and heartbreaking. I grinned most of the way through Mutual Appreciation, so that deserves a mention as well.

Among traditional comedies, The 40-Year Old Virgin was consistently funny without being overly mean, an all-too-rare combination, and so that gets my vote here.
